摘要
目的探讨子痫前期患者胎盘组织α2-HS-糖蛋白(AHSG)基因mRNA及蛋白表达与胰岛素抵抗的关系。方法检测36例正常足月妊娠妇女(A组)和24例轻度子痫前期患者(B组)及17例重度子痫前期患者(C组)剖宫产前空腹血及新生儿脐血AHSG、血糖、胰岛素水平,采用RT-PCR、Western blot法测定胎盘组织中AHSG基因mRNA及蛋白表达水平。结果B、C组新生儿脐血AHSG水平均显著高于A组(P<0.01)。C组新生儿脐血胰岛素、血糖水平显著低于A组(P<0.05)。A、B、C三组胎盘组织中AHSG基因mRNA表达水平分别为0.57±0.17、0.64±0.14、1.32±0.38,C组显著高于B、A组(P<0.05)。A、B、C三组胎盘组织中AHSG蛋白表达水平分别为0.28±0.11、0.30±0.10、0.58±0.10,C组显著高于A组和B组(P<0.05)。结论子痫前期患者新生儿脐血AHSG水平显著高于正常足月妊娠组新生儿。重度子痫前期患者胎盘组织中AHSG基因mRNA及蛋白表达水平显著增高,胰岛素抵抗在子痫前期发病中有重要作用。
Objective To study the relationship between expressions of α2-HS-glyeoprotein gene(AHSG) mRNA and AHSG protein in placenta and insulin resistance in preeclampsia patients. Methods Fasting maternal serum and umbilical cord blood AHSG, insulin and blood glucose(Glu) levels were detected in 36 cases of normal pregnancy woman(group A), 24 cases of mild preeclampsia (group B) and 17 cases of severe preeclampsia (group C). The expressions of AHSG; mRNA and AHSG protein in placenta were detected by semi-quantitative RT-PCR and Western blotting. Results The levels of AHSG in mbilical cord serum were significantly higher in group B and C than those in group A (P〈0. 01). The levels of umbilical cord serum Glu and insulin were significanhy lower in group C than those in group A (P〈0. 05). Expression of AHSG mRNA in placenta of group C (1.32 ±0. 38) was significantly higher than that in group A (0. 57±0. 17)or group B(0. 64±0. 14) (P〈 0. 05). Expression of AHSG protein in placenta of group C(0. 58±10) was significantly higher than that in group A(0. 28±0.11) or group B(0. 30±0. 10) (P〈0. 05). Conclusion The level of AHSG in umbilical cord blood is significantly higher in preeclampsia neonate. Overxpression of AHSG mRNA in placenta of severe preeclampsia patients may induce insulin resistance, which may play an important role in the pathogenesis and development of preeelampsia.
