摘要
目的通过DNA-PKCS反义寡核苷酸转染肺癌细胞A549,探讨DNA-PKCS对肺癌细胞放射敏感性影响。方法设0、0.5、1.0、2.0、4.0、6.0、8.0Gy放射剂量点处理对照组(与转染无关的寡核苷酸)和实验组(转染DNA-PKCS反义寡核苷酸),用成克隆法分析细胞存活分分数,并分别用线性二次模型和多靶单击模型拟合出细胞存活曲线,求出放射生物学参数d、13、SF2、D0、Dq和N,评价细胞放射敏感性变化。结果对照组仪值为0.14,p值为0.030,SF2值为0.63,D0值为2.38,Dq值为1.43。实验组α值为0.31,β值为0.018,SF2值为0.41,D0值为2.09,Dq值为0.60。实验组细胞的α值增大,β、SF2、D0、Dq值均减少。结论DNA-PKCS反义寡核苷酸可增强A549细胞放射敏感性,DNA-PKCS是治疗肺癌的潜在靶点。
Objective To observe the effect of DNA-PKCS antisense oligodeoxynucleotides (ASODN) on the radiosensitivity of lung cancer cell line A549 through transfeeting DNA-PKcs ASODN into A549 cells. Methods DNA-PKCS ASODN and unrelated ODN were transfeeted into A549 cells (testing group and control group). These cells were irradiated with 0,0.5,1.0,2.0,4.0,6.0 or 8.0 Gy X-ray. Clonogenie assay was performed to determine the survival fraction. The parameters D0, Dq and N for the multitarget single-hit model, as well as the parameters α, β and SF2 for the linear-quadratic model,were calculated to evaluate cell radiosensitivity. Results In the control group and testing group,the et value was 0.14 and 0.31 ,The 13 value was 0. 030 and 0. 018 ,the SF2 was 0.63 and 0.41 ,the Do was 2.38 and 2.09 ,the Dq was 1.43 and 0. 60, respectively. In the testing group, the α value of A549 cells was increased, but the β value, SF2, Do and Dq were decreased. Conclusions DNA-PKCS ASODN can enhance the radiosensitivity of A549 cells and is a potential target in treating lung cancer.
