摘要
目的研究广东地区汉族人群人类血小板特异性抗原HPA-15(Gov)系统多态性。方法用PCR-SSP方法对广州血液中心随机采集的500名汉族无偿献血者进行HPA-15系统基因分型。结果广东地区500名汉族无偿献血者HPA-15aa,HPA-15bb和HPA-15ab的基因型频率分别为25%、21%和54%。研究人群HPA-15的基因型分布与Hardy-Weinberg(HW)平衡法则吻合。与其他4个省份汉族人群中HPA-15基因频率的分布比较无显著性差异(P>0.05);在与其他种族人群HPA-15分布的比较,与南美洲的印地安人和非洲的贝宁人群有显著性差异(P<0.05)。结论HPA-15系统具有多态性,且HPA抗原分布不配合比率较高,这对研究同种免疫血小板减少症和开展血小板同型输注的相关研究提供实验基础。
Objective To investigate the polymorphism of the human platelet antigen HPA-15 (Gov) system in Guangdong Han population. Methods A total of 500 unrelated Chinese Han blood donors from Guangdong province of China were genotyped for HPA-15 system using PCRSSP assay. Results Genotype frequencies of HPA-15aa, HPA-15bb and HPA-15ab were 25%, 21% and 54%,respectively. The distributions of HPA-15 in 500 Chinese fit Hardy-Weinberg equilibrium. The distributions of the HPA-15 alleles were no significantly difference among the Chinese in the investigated four provinces(P〉0.05). In comparison to other ethnic groups, signif- icant differences were observed in Xikrin Amerindians and Beninese people(P〈0.05). Conclusion HPA-15 system has polymorphism and the chance of HPA-15 antigen incompatibility in random platelet transfusion is quite high, which provide a basis for further study on alloimmune thrombocytopenic syndromes and HPA-matched transfusion.
