摘要
目的:获取鮟鱇鱼皮中硫酸皮肤素的二糖结构信息。方法:以硫酸软骨素ABC酶酶解硫酸皮肤素样品,得到二糖;对酶解时间进行优化,进而以毛细管液相色谱法和毛细管液相-质谱联用法分析二糖结构及含量。结果:鮟鱇鱼皮硫酸皮肤素在硫酸软骨素ABC酶作用下,37℃反应4h,可酶解完全。其糖链是以艾杜糖醛酸-[1→3]-N-乙酰氨基半乳糖-4硫酸(△Di-4S(Ido))为主要二糖重复单位,此外,还含有少量的艾杜糖醛酸-[1→3]-N-乙酰氨基半乳糖-6硫酸(△Di-6S(Ido))和葡萄糖醛酸-[1→3]-N-乙酰氨基半乳糖(△Di-0S)二糖单位。△Di-4S(Ido)、△Di-0S与△Di-6S(Ido)3种不饱和二糖的摩尔比为171∶1∶1。
Objective: To obtain the information of the disaccharides in dermatan sulfate from Lophius litulon skin. Method: The DS sample was treated with chondroitinase ABC to afford disacchafides and the digesting time was optimized. CLC and CLC-MS method was used to analysis and determinate the disaccharide units content and structure in DS fragment. Result: The sample can be well digested at 37 ℃ for 4 hours treated with chondroitinase ABC. Polysaccha- ride chains of the DS consists of a prevailing disaccharide unit IdUA-[1→3]-GalNAc-4S(△Di-4S(Ido)) and some IdU- A-[1→3]-GalNAc-6S(ADi-6S(Ido)) and GlcUA-[1→3]-GalNAc(ADi-OS), their mole ratio is 1.71:0.01:0.01.
