孕妇不同铁浓度下淋巴细胞增殖反应及TNF-α、IL-10分泌水平

A study on proliferation of lymphocytes and cytokine production of TNF-α and IL-10 of pregnant women under different iron concentrations.

摘要 目的比较不同铁浓度下孕妇淋巴细胞增殖功能以及Th1/Th2细胞因子分泌水平,为临床合理补铁提供理论依据。方法孕妇血肝素抗凝,分离淋巴细胞。给予PHA刺激后,与铁浓度分别为5μmol/L、25μmol/L、50μmol/L的细胞培养液混合培养,MTT比色法检测缺铁性贫血孕妇及健康孕妇淋巴细胞的增殖反应;酶联免疫吸附法(ELISA)检测贫血孕妇淋巴细胞Th1/Th2细胞因子的分泌水平。结果体外试验显示:1.贫血孕妇各组淋巴细胞刺激指数低于健康孕妇(P<0.001);健康孕妇不同铁浓度组淋巴细胞刺激指数有显著性差异(P<0.001)。2.不同铁浓度组贫血孕妇淋巴细胞Th1/Th2细胞因子的分泌水平有显著性差异(P<0.001)。结论孕妇铁营养缺乏或补充过剩都会导致淋巴细胞增殖能力以及细胞亚群功能的下降,因此,孕期应加强含铁食物的摄取,重视铁营养水平的监测,避免因铁营养失衡造成的免疫功能的下降。 Objective: To explore the proliferation of lymphocytes and cytokine production of Th1/Th2 cells of pregnant women under different iron concentrations. Methods : Lymphocytes were isolated from pregnant women and cultured with PHA under different iron concentrations. The proliferation of lymphocytes was tested with MTr and cytokine production of Th1/Th2 cells was tested with ELISA kit specific for human cytokines. Results: In vitro study showed: I. A significant decrease in the proliferation of lymphocytes was observered in iron deficient pregnant women. Addition of iron to the culture medium did not affect the lymphocyte proliferations of iron deficient pregnant women, but caused a change in that of health pregnant women. 2. There was a significant difference in cytokine production of Th1/Th2 cells of iron deficient pregnant women under different iron concentrations. Conclusion: Iron deficiency or overload will result in the decrease of the proliferation of lymphocytes and cytokine production of Th1/Th2 cells of pregnant women. The iron concentration should be monitor in time to avoid decrease of proliferation of lymphocytes and cytokine production of Th1/Th2 cells.
