斑点免疫金渗滤法同时检测猪瘟和猪繁殖与呼吸综合征抗体的研究

Studies on Dot Immunogold Filtration Assay for Detection of Antibody of Hog Cholera Virus and Porcine Reproductive and Respiratory Syndrome Simultaneously

在猪瘟、猪繁殖与呼吸综合征单重斑点免疫金渗滤法的基础上,建立能同时检测HC和PRRS的双重斑点免疫金渗滤法。将提纯的HC和PRRS抗原同时包被在硝酸纤维膜上,利用胶体金标记SPA显色,当猪血清中含有HC、PRRS抗体时,则相应的点会显现红色(呈红色斑点),结果易于判断。且操作简单,耗时短,与猪细小病毒、口蹄疫和猪伪狂犬病毒阳性血清不发生交叉反应。

On the basis of single dot immunogold filtration assay （sDIGFA）for detection of antibody of HC or PRRS,double DIGFA which could detect HC and PRRS antigen simultaneously was established. The purified antigens of HCV and PRRS were coated on nitrocellulose （NC）membrane,and staphylococcal protein A （SPA）labelled with colloidal gold was used for reaction reagent. As swine sera contain both HC and PRRS antibodies,the color of the positive dots （red dots）were so bright that the results could be easily and quickly determined. Furthermore,this method had a better specificity without cross-reaction with positive sera to PPV,FMD and PRV.
