摘要
为满足人工栽培对雨久花种苗的大量需求,以组织培养及无性系建立的方法进行了雨久花快速繁殖的研究。试验以雨久花的嫩茎为材料,进行了愈伤组织的诱导、分化、不定芽生根、试管苗移栽与移植的研究。结果证明,MS+BA 0.6 mg.L-1+2,4-D 1.2 mg.L-1是诱导嫩茎形成具有分化能力愈伤组织的理想培养基,1/2MS+Ag(N03)20.6 mg.L-1是诱导愈伤组织分化的理想培养基,1/3MS+IAA0.5 mg.L-1是试管苗生根培养的理想培养基。试管苗在温室中移栽成活率可达98%以上,移植于池塘边后试管苗生长旺盛。建立起的雨久花无性系可作为人工栽培的种苗。
In order to meet the increasing demand for more seedlings of Monochoria korsakowii Regel et Maack in practice, rapid propagation was carried out by tissue culture and clone establishment. The young stem of Monochoria korsakowii Regel et Maack was used as the experimental material for callus induction, differentiation, rooting from uncertain buds and transplanting of test tube seedlings. The results showed that the most favorable medium for inducing the young stem to form callus is MS + BA0.6 mg.L^-1 +2,4 -D 1.2 mg.L^-1, the most favorable medium for callus differentiation 1/2MS + Ag( NO3 )20.6 mg .L^-1, the most favorable medium for root growth 1/3MS + IAA0.5 mg . L^-1. The survival ratio for plantlet transplanting in greenhouse is more than 98%. The plantlets transplanted in the edge of pond grow vigorously. The clones of Monochoria korsakowii can be used as seedlings in cultivation.
