摘要
目的:探讨肿瘤坏死因子-α(TNF-α)在大鼠移植肾缺血预适应(IPC)中表达及作用。方法:36只成年Sprague-Dawley(SD)大鼠为供受体,并随机分为以下3组:假手术组,剖腹探查后关腹;普通移植组,同种原位肾移植;缺血预适应组,供肾15min缺血10min灌注预处理肾移植。术后24h,应用生化和组织学指标检测各组肾脏功能与肾组织损伤程度,分别用逆转录聚合酶链式反应(RT-PCR)技术和酶联免疫吸附法(ELISA)检测肾组织及血清中TNF-α转录与表达水平。结果:肾移植后24h与普通移植组组相比,缺血预适应组肾小管损伤程度明显减轻(肾小管损伤指数P<0.01),TNF-α转录与表达亦明显减弱(P<0.01);普通移植组和缺血预适应组Scr、BUN水平较假手术组明显升高,但两组间未有明显差异(P>0.05)。结论:用15min/10min缺血预适应能够改善大鼠移植肾早期缺血再灌注损伤,TNF-α表达被抑制可能在预缺血再灌注保护中起着关键性作用。
Objective:To investigate the expression and role of TNF-α on rat renal graft administered with ischemic preconditioning. Methods: Thirty-six male SD rats were served as donors and recipients. Animals were randomly divided into the following 3 group: the sham-operated group (group A), common transplantation group (group B) and treatment group (group C). Animals in A group were subjected to exploratory laparotomy. Group B received orthotopic homotransplantations. IPC in C group was administered with a 15 minutes ischemic followed by a 10 minutes reperfusion. The levels of Scr, BUN, the degree of iscbemic reperfusion injury of kidney graft were assessed. TNF-α levels in serum and graft tissue were detected by RT-PCR and enzyme-linked immunosorbent assay (ELISA), respectively. Results: The degree of renal graft tubules injury in C group was significantly lower compared with the Group B ( P〈0.01 ) as well as the transcription and expression of TNF-α(P〈0.01 ) after 24 hours of transplant. The levels of Scr, BUN were elevated than the sham operated group, whereas no significant difference between Group C and Group B (P〉0.05). Conclusions: One cycle schedule of preconditioning attenuated rats renal graft ischemia reperfusion injury in early phase. The inhibition of expression of TNF-α may play key role for the protection.
