摘要
目的研究甲状腺素对大鼠垂体-卵巢轴内分泌功能及作用机制。方法采用放射免疫测定法,观察甲状腺素不同含量对大鼠血清黄体生成素(LH)、卵巢刺激素(FSH)、雌二醇(E2)、孕酮(P)、肿瘤坏死因子(TNF)、白细胞介素-1β(IL-1β)水平的影响。结果左旋甲状腺素钠(T4)0.1mg/(kg·d)可显著降低大鼠血清LH水平(P〈0.01),而FSH、E2、P、TNF、IL-1β含量均未受明显影响(P〉0.05)。T4 0.6mg/(kg·d)可使大鼠血清LH、E2、P水平明显降低(P〈0.01),而FSH含量变化无统计学意义(P〉0.05),使IL-1β水平显著升高。结论甲状腺素大剂量长期应用对大鼠垂体-卵巢轴内分泌功能均有抑制作用,其机制与细胞因子IL-1β分泌增多有关。
Objective To study the effects of thyroxine(T4) on pituitary-ovary axis in rats and its mechanism. Methods The serum LH, FSH, E2 , P, IL-1βand TNF concentration was measured with radioimmunoassay (RIA). Results T4 [ 0.1 mg( kg. d), ig, 1 d ] decreased serum LH concentration in rats significantly ( P 〈 0.01 ), but had no effect significantly on serum FSH, E2,P,TNF, IL-1βlevel. 0.6 mg(kg .d) T4 decreased serum LH, E2 ,P concentration while increased serum IL-1 βconcentration significantly in rats(P 〈 0.01 ). Conclusion T4 had an inhibiting effect on endocrine function of pituitary-ovary axis in rats by large dose T4 and using for long time. The mechanism is related to the increasing of IL-1 secretion in rats.
