摘要
目的探讨益生元对胃癌细胞的作用效果,为开发安全、有效的预防和辅助治疗胃癌的药物奠定基础,并为益生元的有效应用打开新思路。方法用MTT法研究不同浓度大豆低聚糖、低聚木糖、低聚果糖和低聚甘露糖在不同时间对BGC-823细胞的抑制作用。结果大豆低聚糖、低聚木糖、低聚果糖和低聚甘露糖4种益生元对BGC-823细胞均有一定程度的抑制作用(P<0.05),抑制效果基本呈浓度时间依赖关系,但大豆低聚糖、低聚果糖的作用在48h和72h差别不大。在这4种益生元中,大豆低聚糖的抑制效果最强,低聚木糖次之,低聚果糖、低聚甘露糖的抑制效果较弱。作用48h、72h达到对BGC-823细胞半数抑制率的大豆低聚糖的浓度是100mg/ml左右;作用72h时达到对BGC-823细胞半数抑制率的低聚木糖浓度在100~125mg/ml;而125mg/ml低聚果糖和低聚甘露糖处理胃癌BGC-823细胞72h,均未达到50%的抑制率。结论大豆低聚糖、低聚木糖、低聚果糖和低聚甘露糖对人胃癌细胞株BGC-823细胞有一定程度的抑制作用。
Objective To provide a basis for the study on the inhibition function of prebiotics to Gastric cancer and for the development of safe, effective and less cosdy medicine as the curative or assistant curative to Gastric cancer,and to provide a new idea for more effective application of prebiotics. Method MTT-method was used to investigate the cell survival rate of BGC-823 in different concentrations of Prebioties at different time. Results The inhibition of different of concentrations soybean oligosaccharide, Xylo-oligo-saccharides, Fructo-oligosaccharides and Man-oligosaccharides to the BGC-823 at different times indicated that : ( 1 ) all the prebiotics at certain concentration had an inhibition effect to the growth of BGC-823, which is time and dose dependent (P 〈 0.05 ) ;(2 ) there was no significant difference between the inhibition effect of soybean oligosaccharide and Fructo-oligosaccharides at 24 h and 48 h. Among these four prebiotics,soybean oligosaccharide had the most obvious effect ,followed by Xylo-oligosaccharides, while Fructo-oligosaccharides and Man-oligosaccharides hsd the least effect than them. When the reaction time was 48 h or 72 h,the half inhibitory concentration of soybean oligosaccharide was about 100 mg/ml; When the reaction time was 72 h, the half inhibitory concentration of Xylo-oligosaccharides was between 100 mg/ml and 125 mg/ml. The inhibition rates of Fructo-oligosaccharides and Man-oligosaccharides failed to reach 50% even in 125 mg/ml for 72 h. Conclusion Soybean oligosaccharide, Xylo-oligosaccharides, Fructo-oligosacchariides and Man-oligosaccharides have evident inhibition effect on proliferation of BGC-823 in vitro.
