峨边花牛肌生长抑制素基因编码序列的克隆与分析

摘要 以乐山峨边花牛骨骼肌为试验材料,提取总RNA为模板,利用RT-PCR技术和常规T-A克隆法,通过PCR鉴定及序列测定,将所测得的编码序列与已发表的编码序列同源性分析比较,发现没有出现引起氨基酸改变的突变。结果表明:乐山峨边花牛肌生长抑制素基因仍很完整,可通过改变基因结构等来降低肌生长抑制素活性的方法改良地方黄牛品种并提高产肉率。 In this study,the Myostatin gene was amplified by RT- PCR from the total RNA of Ebian spotted cattle skeletal muscle. Then it was cloned into vector by T - A cloning method. According to the PCR identification and sequence analysis,it indicated that the Myostatin gene of Ebian spotted cattle was cloned successfully by sequeeing. This has laid a solid foundation for its expression in eukaryotic and prokaryotic vector and provides a new way to improve the native yellow cattle breeds and meat production rate.
