RNA干扰沉默COX-2对鼻咽癌细胞增殖、凋亡及细胞周期的影响 被引量:5

摘要 目的筛选COX-2基因的有效RNA干扰序列,研究沉默COX-2表达对鼻咽癌细胞增殖、凋亡以及细胞周期的影响。方法人工合成双链的小干扰RNA(siRNA),脂质体法转染鼻咽癌C666-1细胞后,RT-PCR法检测COX-2的mRNA含量表达变化,筛选出沉默COX-2表达的有效RNA干扰序列。MTT法检测细胞增殖的变化,DAPI染色检测细胞凋亡,流式细胞法进行细胞周期分析。结果设计合成的anti-COX-2b siRNA能够使COX-2mRNA表达下降90%以上,COX-2基因沉默后,鼻咽癌细胞周期出现G0/G1期阻滞,增殖能力下降,但对凋亡无明显影响。结论RNA干扰沉默COX-2的表达能够抑制细胞增殖,阻滞细胞周期,可以成为鼻咽癌研究和抗肿瘤基因治疗提供新的思路,进行深入研究。
