中华眼镜蛇毒神经生长因子对PC12细胞κ阿片受体蛋白质表达的影响

Effects of nerve growth factor from cobra(Naja Naja Atra) venom on expressions of κ opioid receptor protein in PC12 cells

目的探讨中华眼镜蛇毒(Naja Naja Atra)神经生长因子(nNGF)对PC12细胞κ阿片受体蛋白质表达的影响。方法PC12细胞分为正常对照组和给药组,采用Western blot方法检测各组PC12细胞κ阿片受体蛋白质的表达。结果①nNGF的25、50、100μg.L-13个剂量中,50和100μg.L-1nNGF下调PC12细胞κ阿片受体蛋白质的表达,50μg.L-1剂量组下调最明显(P<0.01),100μg.L-1剂量组下调(P<0.05)。②50μg.L-1nNGF分别处理PC12细胞1、3、5、7、10 d后,其κ阿片受体蛋白质表达呈降低趋势,其中d 5、7下调(P<0.05),d 10下调明显(P<0.01)。③10μmol.L-1吗啡可上调PC12细胞κ阿片受体蛋白表达,而10μmol.L-1纳络酮对其无影响,二者分别都能逆转nNGF的下调效应,但是二者共存时不能逆转nNGF的下调效应。结论nNGF诱导PC12细胞分化中,可引起PC12细胞κ阿片受体蛋白质表达的下调效应,该效应能被吗啡或纳络酮逆转,但二者共存不能被逆转效应。

Aim To examine the effects of nerve growth factor from cobra （Naja Naja Atra ） venom （nNGF） on expressions of K opioid receptor protein in PC12 cells. Method Rats were divided into normal control group and experimental group to measure the effects of nNGF on expressions of K opioid receptor protein in PC12 cells by Western blot. Results （1） 50μg·L-1 nNGF most significantly （P 〈0.01 ） or 100μg·L-1 nNGF （ P 〈 0.05 ） significantly down-regulated expressions of K opioid receptor protein among the three dosages of 25,50 and 100 μg·L-1 . （2） Expressions of K opioid receptor protein in PC12 cells displayed down-regulation within 10 days after PC12 cells being added with 50 μg·L-1 nNGF and were downregulated significantly in the 5 th and 7 th day （P 〈 0.05 ） and in the 10 th day the most significantly （P 〈 0. 01）. （3） 10 μg·L-1 morphine up-regulated ex- pressions of K opioid receptor proteins in PC12 cells but 10 μg·L-1 naloxone did not. They could re- verse respectively the down-regulation of their expres- sions induced by nNGF, but they together could not. Conclusions When nNGF induced the differentiation of PC12 cells, expressions of endogenous K opioid receptor proteins in PC12 cells were down-regulaed, and this down-regulation was reversed by morphine or naloxone,but not by them together.
