人α_1抗胰蛋白酶(α_1-AT)基因cDNA的克隆与序列分析

摘要 目的:克隆人α1抗胰蛋白酶基因的cDNA并进行序列分析。方法:以人胚肾细胞系HEK293的mR-NA为模板,采用RT-PCR方法得到人α1-AT基因的cDNA,克隆至载体pUC19中,酶切鉴定后进行序列分析。结果:获得人α1-AT基因的cDNA和重组质粒pUC19-AT,DNA序列分析证实获得的α1-AT基因cDNA,其序列和文献报道一致。结论:采用基因克隆的方法得到人α1-AT基因cDNA,为进一步进行其结构和功能研究打下了基础。 Objective: To clone the gene encoding human alphal -antitrypsin (α1-AT) . Methods:Using the isolated total RNA from human embryo kidney cell line HEK293 as the template, the cDNA encoding cq - AT was amplified by reverse transcription - polymerase chain reaction ( RT - PCR) method, and then cloned into pUC19 - AT and sequenced. Results: The cloned human α1 - AT sequence was the same as reported. Conclusion: The geng encoding the human α1-AT successfully acquired by gene cloning, which have made a foundation for its further functional research.
