摘要 目的:探讨缺氧诱导因子(HIF-1α)和P300/CBP在肺鳞癌和肺腺癌中的表达及其与分型、分化、转移的关系。方法:采用免疫组织化学(SP)法检测60例肺鳞癌和肺腺癌组织中HIF-1α及P300/CBP的表达。结果:HIF-1α蛋白表达的阳性率在癌组织中为48.33%,显著高于正常支气管黏膜组织(P<0.01),P300/CBP的表达与肿瘤的分化程度、淋巴结转移和患者吸烟与否显著相关,但与年龄性别无关,肺鳞癌和肺腺癌组织中HIF-1α表达与P300/CBP表达呈显著负相关(P<0.01)。结论:肺鳞癌和肺腺癌组织中HIF-1α蛋白表达与靶蛋白P300/CBP蛋白降解有关,提示HIF-1α蛋白过表达在肺癌的发生和发展中起重要作用。 Objective:To investigate the expression of HIF -lct and P300/CBP in non - small cell lung cancer and to find out their correlation with histological type, cell differentiation, metastasis of squamous cell carcinoma and of lung. Methods:The expression of HIF - 1α and P300/CBP was detected in 60 NSCLC tissues by highsensitive S - P immunohistochemical method. Results:The abnormal expression rate of HIF - 1α was 48.33% in cancer tissues. Expression of HIF -1α protein was detected in a significantly greater proportion in squamous cell car- cinoma and adenocarcinoma of lung than that in normal bronchial epithelium ( P 〈 0.01 ) ; There was no relationship between HIF- lot expression and age or sex; otherwise, there was closely relationship between HIF-1α expression and cellular differentiation, lymph node metastases and smoking; Expression of HIF -1α was negatively correlated with expression of P300/CBP (P 〈 0.01 ). Conclusion:Expression of HIF - 1α protein reduced the protein level of P300/CBP,indicating HIF- 1α plays an important role in oncogenesis and development of lung cacncer.
