A new model for laser-induced choroidal neovascularization in rabbits

摘要 目的评价半导体泵浦的倍频Nd:YAG532nm绿激光诱导色素兔脉络膜新生血管(choroidal neovascularization,CNV)模型的可行性。方法将24只色素兔按光凝后1周、2周、3周、4周分为4组,每组6只。用高强度倍频Nd:YAG532nm绿激光光凝色素兔视网膜,分别于激光照射后第1、第2、第3及第4周行荧光素眼底血管造影(fundus fluorescein angiography,FFA),FFA检查后处死动物,取光凝区眼球壁制作标本,进行光镜和透射电镜观察及CD34免疫组织化学染色,以观察激光诱导CNV的效果。结果光凝后第1周,FFA检查见脉络膜期及动脉早期10.0%的光斑有点状荧光素渗漏;光凝后第2、第3、第4周,依次有37.5%、51.7%、45.0%的光斑出现脉络膜新生血管形态。病理切片及CD34免疫组化染色证实,光凝后第1、第2、第3、第4周,兔CNV发生率分别为33.3%、57.1%、66.7%及61.5%。结论通过半导体泵浦的倍频Nd:YAG532nm绿激光诱导色素兔CNV模型是可行的。 Objective To create a pigmented rabbit model of choroidal neovascularization (CNV) with a frequency-doubling Nd: YAG 532 nm pumping diode laser. Methods Twenty-four pig- mented rabbits were randomly divided into 4 groups by every schedule point at 7 days interval. One week, 2 weeks, 3 weeks and 4 weeks after photoeoagulation with a high-energy Nd:YAG 532 nm laser, the effect of the laser on inducing CNV was evalu- ated by fundus fluorescein argiography (FFA), light microscopy, transmission electronmicroscopy and the immunohistochemical analy- sis of CD34. Results The leakage in FFA associated with CNV was first detected by 1 week after photocoagulation (12/120 lesions, 10.0%); by 2, 3 and 4 weeks after photocoagulation, evidence of choroidal neovascularization (CNV) appeared on 37.5%, 51.7%, and 45.0% of the spot, respectively. Histological sections proved the incidence of CNV was 33.3%, 57.1%, 66.7%, and 61.5% at 1, 2, 3, and 4 weeks after photocoagulation. Conclusion It is f easible to create a pigmented rabbit model of CNV with a frequency-doubling Nd:YAG 532 nm pumping diode laser.
