环境污染水对C57BL/6J小鼠繁殖发育的毒性影响

摘要 目的:考察工业和生活排污水中可疑卤代芳烃化合物的污染对C57BL/6J小鼠繁殖和发育的影响。方法:成年雌性小鼠40只,随机分4组,每组10只,Ⅰ组:饮用自来水作为正常对照组;Ⅱ组:饮用浓度2ng/L2,3,7,8-四氯苯对二英(TCDD)标准水为阳性对照组;Ⅲ组:饮用实验1号污染水;Ⅳ组:饮用实验2号污染水。连续繁殖3代,观察两种污染水对小鼠繁殖和发育的影响。结果:Ⅱ组和Ⅲ组的亲代(P2)受孕率分别为(84.62±6.08)%、(87.50±33.61)%,比正常对照组下降(P<0.05);Ⅱ、Ⅲ和Ⅳ组的亲代(P2)活产率均下降,分别为(80.00±40.00)%、(57.14±49.49)%、(58.06±49.35)%,与正常对照组比较差异有统计学意义(P<0.01)。Ⅱ组和Ⅳ组子代(F3)离奶体重降低为(8.59±1.59)g、(8.94±1.12)g,与正常对照组比较差异有统计学意义(P<0.001);Ⅱ组、Ⅲ组和Ⅳ组子F2代和F3代离奶成活率均下降,分别为(50.21±48.14)%、(75.34±40.22)%、(61.48±42.09)%和(41.33±40.66)%、(74.38±43.03)%、(74.61±39.53)%,与正常对照组比较差异有统计学意义(P<0.01)。结论:广州部分工业和生活废水有低剂量卤代芳烃化合物污染,长期接触将影响雌性小鼠生殖功能,造成受孕率和活产率的下降、出现流产和初生后子鼠死亡率升高、断乳成活率和重量的降低。 Objective: To evaluate the toxic effects of waste water suspected to contain arylcontaminants on reproduction and development of mice. Methods:Forty female mice were randomly divided into 4 groups (n=10 each) on normal drinking water as controls (group Ⅰ ), or 2 ng/L TCDD water (group Ⅱ), or No.1 sample water (group Ⅲ) or No.2 sample water (group Ⅳ). The mice were allowed to pass on three consecutive generations, and were studied for general toxicity (fertility, birth rate, and weanling survival and weight of the three generations). Results: The results showed significantly reductions in fertility of the groups Ⅱ and Ⅲ [(84.62±6.08)%, (87.50± 33.61)%, P〈0.05] compared with group Ⅰ in the generation P2, in birth rates of group Ⅱ, group Ⅲ and group Ⅳ [(80.00±40.00)%, (57.14±49.49)%, (58.06±49.35)%, P〈0.01 ] compared with the control group (P〈0.01) in the generation P2, in weanling weights of group Ⅱ and group Ⅳ [(8.59± 1.59)g, (8.94± 1.12)g, P〈 0.001)] compared with the control group Ⅰ in generation F3, in weanling survival of group Ⅱ, group Ⅲ and group Ⅳ [(50.21±48.14)%, (75.34±40.22)%, (61.48± 42.09)%, (41.33±40.66)%, (74.38±43.03)%, (74.61±39.53)%, P〈0.01 ] compared with the group Ⅰ in generation F2 and F3. Conclusion: Long-term exposure to low-dose aryl-contaminants in waste water may be lead to harmful effects on fertility, reduced rate of pregnancy, lower live birth in mice.
