摘要
目的:探讨复方水蛭素提取物(COSZSEX)对小鼠移植性肿瘤和th1/th2类细胞因子的作用机制,为临床用药提供理论依据。方法:1)COSZSEX体内抑瘤实验:取小鼠40只,随机分组,均接种S180瘤细胞。阴性对照组给予生理盐水,阳性对照组给予5-FU,实验组分别给予大小剂量的COSZSEX,连续用药14 d,比较各组肿瘤生长情况,并计算抑瘤率。2)检测th1/th2类细胞因子:取荷瘤鼠组织标本,提取细胞总RNA,紫外分光光度计测出A260及A280值。逆转录(RT)反应,聚合酶链反应(PCR),电泳鉴定。将凝胶电泳图像输入美国Kodak凝胶分析系统,应用1D I mage Analysis Software进行表达强度分析。结果:COSZSEX对小鼠移植性肿瘤具有明显的抑制作用,大小剂量组抑制率分别为49.06%和35.53%,大剂量组优于小剂量组。荷瘤鼠用药后,IL-2和IFN-γ细胞因子升高,IL-10和IL-4细胞因子降低。结论:COSZSEX对小鼠移植性肿瘤具有显著的影响,同时使荷瘤鼠低下的Th1类细胞因子增多,提高了免疫应答能力。
OBJECTIVE: To disscuss the mechanism of COSZSEX on mouse's trsnsplantable tumor and CK th1/th2, so as to pro-ide the evidence for application in clinic. METHODS: 1)The experiment of restrain tumor in body of COSZSEX: 40 mice were randomed to a negative comparison group and opositive eomparision group, and all the mice were inoculated with S180 tumor cells. Saline, and 5-FU were used respectively in negative comparison group and positive corn parison group. COSZSEX which was divided into big dose and small dose was given to the treatment groups for 14 days, The growth of rumors of the different groups was observed and compared, and the rates of restrain tumors were caculated. 2. CK-th1/th2: The cell's to tal RNAs from the bearing cancer mouse's tissue samples were extracted and A260 and A280 were measured by an ultraspectrophotometer. RT,PCR, and electrophpresis identity were performed. The GE pictures were put into American Kodak Gel analytical system, and ID Image Analysis Software was used to express the intensity analysis. RESULT: COSZSEX effectly restrained the mouse's transplantable tumor, the restrain rates were 49.06% and 35.3% rspectively, and the big dose group was better than the small dose group. After the bearing cancer mouse took the medicine, CK-IL-2 and CK-IFN-r increased and CK-IL- 10 and CK-IL-4 decreased. CONCLUSIONS: COSZSEX has an obvious effect on the mouse's transplantable tumor, and at the same time, it makes CK thl which is low in the bearing cancer mouse's body increase, the immunity improve and the imbalancable CK-th1/th2 reache the state of balance.
