摘要
目的检测腹泻模型大鼠在给予洛哌丁胺处理后结肠黏膜水通道蛋白4(AQP4)mRNA表达的变化,探讨洛哌丁胺在结肠水代谢中的分子作用机制。方法采用逆转录聚合酶链反应(RT-PCR)分别对洛哌丁胺治疗组、模型对照组及正常对照组大鼠升、降结肠黏膜细胞AQP4 mRNA表达进行半定量分析。结果①洛哌丁胺治疗组升、降结肠AQP4 mRNA表达量均高于模型对照组及正常对照组(P<0.01)。②模型对照组升结肠AQP4 mRNA表达量较正常对照组大鼠减少(P<0.05),而降结肠表达差异无统计学意义(P>0.05)。结论洛哌丁胺可以在转录水平上调腹泻大鼠结肠黏膜AQP4表达,使结肠对肠腔内水分的吸收增加。
Objective To investigate the effect of loperamide on the expression of AQP4 mRNA in colon mucosa of model rats of diarrhea and to study the eorrelationship between AQP4 expression and loperamide as well as the molecular mechanism for loperamide in colon water metabolism. Methods Semiquantitative reverse transcription-polymerase chain reaction( RT-PCR) was used to detect the colonic mRNA expression of AQP4 in this study. Results AQP4 was expressed in specimens of the proximal and distal colon of three groups. The expression of AQP4 mRNA in loperamide- treated group was increased remarkably than that in model group and control group(P 〈 0.01 ). Its expression was lower in proximal colon of model group than that in control group(P 〈 0.05) , whereas there was no difference in distal colon between model group and control group. Conclusion Loperamide upregulates AQP4 mRNA expression of the proximal and distal colon in model rats of diarrhea. As a result, absorption of water is increased.
