摘要
目的探讨人类β-防御素1(human beta-defensin1,hBD-1)、人类β-防御素2(human beta-defensin2,hBD-2)在声带息肉组织中的表达及意义。方法采用组织芯片技术制作包括声带息肉组、声带小结组以及对照组(正常声带组织)标本的组织芯片,利用S-P免疫组织化学染色法检测hBD-1、hBD-2在各组的表达情况。结果hBD-1在声带息肉、声带小结及对照组中均为低强度表达,且三组间差异无统计学意义(P>0.05);hBD-2在声带小结组和对照组中仅有微弱表达,而在声带息肉组中表达明显增强(P<0.05)。结论hBD-1和hBD-2在声带上皮中分别为固有性表达和诱导性表达,这表明声带上皮在机体细胞和体液免疫尚未发生作用之前,即可通过分泌β-防御素及时、有效地杀灭外来病原菌,在早期炎症反应中发挥天然防御作用。
Objective To investigate the expression of hBD- 1 and hBD 2 in the vocal cord polyp. Methods Tissue chips consisting of vocal cord polyp, vocal nodules and normal vocal cords were made by tissue chip technique. S- P immunohistochemistry was used to determine the expression of hBD- 1 and hBD- 2 in every tissue chip. Results β-defensin 1 was expressed in all tissue specimens at levels that did not differ significantly(P〉0.05). β-defensin 2 was detected in the specimens of vocal cord polyp, but very little in the subjects of other two groups. Its expression level was significantly higher in the vocal cord polyp than that of the other two groups (P〈 0.05). Conclusion These results suggest that beta-defensin of the vocal cord may play an important role in innate defense against microorganisms. It is feasible to detect clinical samples with the tissue chip technology efficiently on a large scale. The future application of the tissue chip technology is very broad.
