摘要 目的:研究丁酸钠诱导结肠癌细胞分化过程中抑癌候选基因NDRG2的表达特点及意义.方法:采用丁酸钠诱导结肠癌HT29,SW480,SW620细胞分化,通过碱性磷酸酶(AKP)测定和透射电镜评价结肠癌细胞的分化程度,应用Realtime RT-PCR和Western Blot法测定不同分化点(0,1,2,3,4,5,6d)NDRG2 mRNA和蛋白的表达特点.结果:经丁酸钠作用24h~6d后,结肠癌细胞中NDRG2基因mRNA和蛋白水平均显著高于对照组水平.结论:丁酸钠诱导结肠癌细胞分化过程中NDRG2表达显著升高,提示NDRG2是参与结肠癌细胞分化的相关基因. AIM: To investigate the effect of sodium butyrate (SB) on the expression of tumor suppressor gene NDRG2 in colon carcinoma cells and its significance. METHODS:Colon carcinoma HT29, SW480, SW620 cells were induced by sodium butyrate. Alkaline phosphatase (AKP) level detection and transmission electron microscope were used to determine the differentiation degree. The expressions of NDRG2mRNA and protein at different differentiation points (0, 1, 2, 3, 4, 5, 6 d) were detected by Real time RT-PCR and Western Blot. RESULTS:The expressions of NDRG2 mRNA and protein were increased significantly by sodium butyrate after induction for 24 h - 6 d ( P 〈 0. 01 ). CONCLUSION: During the differentiation of colon carcinoma ceils induced by sodium butyrate, the expression of NDRG2 increases significantly, indicating that NDRG2 is a gene that participates the differentiation of colon carcinoma cells.
