摘要
目的:观察CXCR4基因转染对大鼠真皮多能干细胞(dermal multipotent stem cells,dMSCs)分化、增殖、迁移等生物学性状的影响。方法:分别采用荧光激活细胞分类法、绘制细胞生长曲线法、细胞培养分化诱导法和Transwell小室培养体系中化学趋化法,观察CXCR4基因转染后,CXCR4阳性细胞数目以及dMSCs增殖、分化和迁移能力的变化情况。结果:CXCR4基因转染后,CXCR4阳性dMSCs数目明显高于对照组(P<0.05),dMSCs增殖能力略高于对照组,但差异无统计学意义(P>0.05);从Transwell小室上层迁移到下层的dMSCs明显高于对照组(P<0.05)。同时,转染CXCR4基因的dMSCs可向成脂、成骨、成肌细胞方向分化。结论:CXCR4基因转染后,dMSCs中CXCR4表达增加,并有效地增强其迁移能力,且未影响dMSCs的增殖和分化能力。
Objective To observe the effect of CXCR4 gene transfection on the differentiation, proliferation, and migration capacity in dermal muhipotent stem cells (dMSCs). Methods The count of CXCR4-positive dMSCs and the proliferation, differentiation, migration capacity of the cells were measured by fluorescence-activated cell sorting, drawing of cell growth curve, cell culture and differentiation induction, and Transwell chemotaxis assay, respectively. Results After CXCR4 gene transfectiou, the count of CXCR4-positive dMSCs was markedly greater than that of the control cells (P 〈 0.05); the proliferative capacity was slightly greater in dMSCs than in the controls, with no statistic significance (P 〉 0.05) ; more CXCR4-positive dMSCs than the controls migrated to the lower chamber from the upper one (P 〈 0.05). The CXCR4- positive dMSCs were capable of differentiating into lipogenie, osteogenic, and myogenic lineages. Conclusion After CXCR4 gene trausfeeion, the expression of CXCR4 on dMSCs is up-regulated, resulting in an enhanced ability of dMSCs to migrate but no changes in the proliferation and differentiation capacity of the cells.
