摘要
为探讨培养基和外植体对杜仲愈伤组织诱导率的影响,将子叶、茎和真叶(代号为B1、B2、B3)三种外植体分别接种于MS、B5、White、Miller、H、(代号分别为A1、A2、A3、A4、A5)5种培养基上,调查并分析各组合的愈伤组织诱导率。经F测验,培养基、外植体和培养基×外植体的互作,效果均极显著。经SSR测验,H、B5、MS三种培养基愈伤组织诱导率极显著高于其它两种培养基。外植体中以茎的诱导率最高。不同组合间以A2B3、A5B3、A5B1、A2B2、A5B2、A1B1、A1B17个组合的愈伤组织诱导率较高,达90%以上。
To select optimal medium and explant for the tissue culture of Ecommia ul-moids, three kinds If explants [cotyledon(B1), stem (B2)and leaf(B3)] were cultured on fivekinds of Media [MS(A1), B5(A2), white(A3), Miller(A1) and H(A5)]. The results indicatedthat medium factor, explant factor and medium × explant factor all had a significant effecton the callus - inducing rate.SSR test was taken to make comparisons among treatment means. the results showedthat callus - inducing rates of H, B5 and MS were all higher than the other two, which wasstatically significant (P< 0.01). Callus - inducing rate of stem explant was the highest,which was statistically significant (P< 0.01). Among all combinations A2B3, A5B3, A5B1,A2B3, A5B2, A2B1and A1B2 combinations were significantly higher than the others.
