摘要
为探讨禽胰多肽(APP)作用的信号传导机制,以Ca3(PO4)2沉淀法制备的APP粗提液及经Sephadex G-50脱盐后层析的APP制品为材料,用不同方式和不同浓度添加APP两种制品进行动物试验,选取90只1周龄艾维茵肉鸡随机分为5组,每组18只。Ⅰ组为对照组,Ⅱ组和Ⅲ组设为饲饮组,Ⅳ组和Ⅴ组为注射组,在第7周末,屠宰取其肝脏和小肠组织,采用放射免疫法(RIA)对其细胞中的环腺苷酸(cAMP)浓度进行测定。结果发现,人为添加或注射APP后,在肝脏组织中各试验组与对照组比较,cAMP浓度变化主要呈下降趋势,但差异不显著(P>0.05);小肠组织中注射组和饲饮APP粗提液组与对照组相比cAMP浓度均下降,并且APP粗提液组极显著降低(P<0.01);而饲饮层析APP制品组与对照组相比呈升高趋势。提示cAMP在APP发挥其功能的信息传递过程中起到了一定作用,APP还可能存在其他的信息转导途径。
To investigate the signal transduction pathway mechanism of avian pancreatic polypeptide(APP), our study was designed as follows: First, crude extract liquid of APP was acquired by calcium phosphate adsorption, after chromatographic fractionation with Sephadex G-50, chromatographic APP was acquired. Afterward, the test with two kinds of APP in animals was done. 90 broilers were randomly grouped into 5 groups and each group has 18. Group Ⅰ was control group, Ⅱ and Ⅲ were fed with APP,Ⅳ and Ⅴ were injected hypodermically with APP. 7 weeks later, they were killed, and the livers, small intestines were gathered, then the concentration of cAMP in the cells were examined by radioimmunoassay (RIA). The results showed that after fed or injected hypodermically with APP, the concentration of cAMP were mostly decreased in livers, but there were no significant differences between test groups and control group (P〉0.05). In small intestines, the concentration of cAMP was decreased in groups injected and fed with crude extract liquid of APP,and the later was significantly different (P〈0.01). Otherwise, the concentration of cAMP was raised in the tests fed with chromatographic APP. The results suggest that cAMP must have some effects on the function of APP,and there must be some other signal transduction pathways of APP.
