摘要
目的探讨天麻钩藤饮对肾血管性高血压大鼠转化生长因子β1(TGF-β1)和胰岛素样生长因子1(IGF-1)的影响。方法取雄性SD大鼠48只,随机分为假手术组、模型组、卡托普利组和天麻钩藤饮组,每组12只。造模后第5周开始给药,连续治疗8周,检测各组大鼠血压、左室重量(LVW)、左室重量指数(LVI=LVW/BW)和心肌组织胶原浓度(Coll),观察天麻钩藤饮抑制心肌纤维化的作用。同时,观察各组大鼠心肌组织转化生长因子β1和胰岛素样生长因子1的表达。结果造模12周后,模型组大鼠的LVW、LVI和Coll均明显高于假手术组(P<0.01),说明模型组大鼠出现了明显的左室肥厚和心肌纤维化现象,卡托普利组和天麻钩藤饮组大鼠的上述指标均有显著降低(P<0.01),说明两药对左室肥厚和心肌纤维化均有明显的抑制作用,而两药之间没有显著性差异(P>0.05)。模型组大鼠心肌组织的TGF-β1水平较假手术组有明显的升高(P<0.01),天麻钩藤饮对TGF-β1有显著的抑制作用(P<0.05),而卡托普利组的TGF-β1水平虽然有所下降,但无统计学意义(P>0.05)。各组间大鼠心肌组织IGF-1水平无显著性差异(P>0.05)。结论天麻钩藤饮能够缓解和逆转心肌纤维化,可能同抑制心肌组织TGF-β1的表达有关。
Objective To observe the effects of Tianma Gouteng Decoction (TGD) on the levels of transforming growth factor (TGF - β1) and insulin -like growth factor (IGF- 1 ) in renovascular hypertensive rats. Methods Forty- eight male SD rats were randomly divided into sham -operation group, model group, eaptopril group and the Tianma Gouteng Decoction group, 12 in each group. From the fifth week of the modeling, the rats were treated with the corresponding drugs for continuous 8 weeks. After treatment, the blood pressure (BP), left ventricular weight (LVW), left ventricular index (LVI) and collagen Ⅱ (Co Ⅱ) were observed in every group to evaluate the inhibitory effect of TGD on myocardial fibrosis. Meanwhile, the levels of TGF - β1 and IGF - 1 in cardiac muscle of every group were also examined to probe into the therapeutic mechanism. Results In 12 weeks of modeling, the LVW, LVI and Coll in the model group were higher than those in the sham - operation group( P 〈 0. 01), indicating the formation of left ventrieular hypertrophy and myocardial fibrosis. The above indexes were decreased in eaptopril and TGD group ( P 〈 0.01 ), but the difference between the two medication groups was insignificant( P 〉 0.05). The level of TGF- β1 in cardiac muscle of the model group was higher than that in the sham- operation group distinctly( P 〈 0.01), and TGD decreased obviously the level of TGF - β1 ( P 〈 0.05). The level of the eaptopril group was decreased, but the difference has no statistical significance( P 〉 0.05) . The difference of IGF - 1 level was insignificant among the three groups ( P 〉 0.05). Conclusion Tianma Gouteng Decoction can reverse left ventricular hypertrophy and myocardial fibrosis, and its mechanism may be related to the inhibition of TGF - β1 expression in cardiac muscle tissue.
