摘要
目的:探讨强肌健力口服液对大鼠肝移植术后受体的白细胞介素-2(IL-2)和白细胞介素-10 (IL-10)的影响。方法:应用二袖套技术建立大鼠原位肝移植模型80只,并将其分为强肌健力口服液(QJJL)组、环孢素A(CsA)组和QJJL+CsA组和未治疗组(对照组)各20只;用双抗体夹心ABC—ELISA法和逆转录多聚酶链反应(PCR)技术,对术前及术后不同时相点采集的各组大鼠外周血的IL-2、IL-10水平及受体肝组织内的IL-2 mRNA、IL-10 mRNA进行检测。对结果进行比较。结果:外周血IL-2、IL-10水平检测显示:QJJL组、CsA组和QJJL+CsA组的IL-2和IL-10的术前含量低于术后各时相,但差异无显著性意义(P>0.05);对照组术后5天血清IL-2的含量明显高于术前和治疗组(P<0.05),IL-10术后5天的含量明显低于术前和各治疗组(P<0.05)。PCR检测显示:各治疗组的IL-2 mRNA的表达明显低于对照组(P<0.01);IL-10 mRNA在各治疗组的表达显著高于对照组(P<0.01)。各治疗组的急性排斥反应的发生率明显低于对照组(P<0.05,P<0.01)。结论:IL-2和IL-10的基因表达与大鼠肝移植术后的免疫反应有关,强肌健力口服液联合免疫抑制剂有助于移植大鼠术后免疫紊乱的防治。
Objective:To observe the effect of Qiangii Jianli Decoction (QJD) on interleukin 2 (IL-2) and interleukin 10 (IL-10) levels in rats after liver transplantation.Methods:Rats models of orthotopic liver transplantation were established by two-cuff technique.The rats were randomized into QJD group,cyclosporine A (CsA) group,QJD + CsA group,and the model con- trol group,20 rats in each group.Enzyme-linked immunosorbent assay (ELISA) with double anti- body sandwich method was used to detect the peripheral blood levels of IL-2 and IL-10,and re- verse transcription-polymerase chain reaction technique was used for the examination of hepatic mRNA expression of IL-2 and IL-10 before transplantation and in different time after transplanta- tion.Results:Except the model control group,the peripheral blood levels of IL-2 and IL-10 were lower before transplantation than those in different time after treatment,the difference being insignificant (P>0.05).Serum IL-2 level was higher and IL-10 level was lower in the control group 5 days after transplantation than that before transplantation and that in the treatment groups (P<0.05).The PCR result showed that IL-2 mRNA expression was lower and IL-10 mRNA expression was higher in the treatment groups than that in the control group (P<0.01).The inci- dence of acute rejection was lower in the treatment group than that in the control group (P<0.05 or P<0.01).Conclusion:The immune disorder in rats after liver transplantation is related with the gene expression of IL-2 and IL-10,and QJD combined with immunosuppressive agents exerts a preventive and curative effect on the immune disorder.
