摘要
目的建立一种改良的急性视网膜坏死动物模型,探讨治疗急性视网膜坏死的治疗方法。方法将不同剂量的单纯疱疹病毒(HSV-1,KOS株)通过不同途径,注射于青紫蓝兔眼内的不同部位,同途径不同剂量注射,比较术后玻璃体混浊程度、视网膜坏死的过程和形态,从而确定操作简便、视网膜坏死灶可以量化的动物模型。结果经玻璃体将病毒直接注射于视网膜表面最简单,但是玻璃体混浊严重,不利于观察视网膜坏死的程度;经后极部巩膜视网膜下穿刺,将病毒注射于视网膜下,可以产生4级的视网膜坏死,且玻璃体较清亮;经玻璃体将病毒注射于视网膜下,视网膜坏死清晰、玻璃体清亮,且操作最简单。8500PFU的HSV-1是合适的接种剂量。结论8500PFU的HSV-1经玻璃体视网膜下病毒接种可以建立良好的急性视网膜坏死动物模型。
Objective To establish an improved animal models of acute retina necrosis. Methods Different dosages of HSV-1 KOS strain were inoculated into varied parts of pigmented rabbits' eyes through different ways, and different dosages through same way. An easygoing and quantified animal model was established by comparing vitreous opacity, the course and extent of retina necrotizing damages. Results Direct inoculation of virus onto the retina surface was the simplest way, but could cause dense vitreous opacity and could not observing the degree of retinal necrosis. Subretinal injection of virus through post-pole sclera could induce four-grade retina damages with relatively clear vitreous except for difficult surgical skills. Inoculation of 8 500 PFU HSV-1 through pars sclera had clear vitreous, satisfied retina damages and was simple. Conclusion Subretinal inoculation of 8 500 PFU HSV-1 can establish a great acute retina necrosis animal model.
