摘要
目的:探讨糖基化终产物(AGEs)对人牙周膜细胞MMP-3及TIMP-1蛋白表达的影响,进一步阐明糖尿病牙周病发病过程中细胞外基质代谢调节的分子机制。方法:将糖基化终产物修饰的人血清白蛋白与人牙周膜细胞在体外共同培养,用免疫荧光法及ELISA技术检测人牙周膜细胞MMP-3、TIMP-1蛋白表达。结果:人牙周膜细胞在体外表达MMP-3,培养3d后,MMP-3表达明显增强,与对照组相比具有显著性差异。而TIMP-1表达减弱,与对照组相比不具有显著性差异。结论:糖基化终产物可显著影响人牙周膜细胞MMP-3的表达,为AGEs参与糖尿病牙周病发病机制提供了直接证据。
Objective:To investigate the effects of the mechanical AGEs on the expression of MMP-3 and TIMP-1 in human periodontal ligament cells (HPDLC), and to unveil the molecular mechanism of ECM metabolism during periodontitis in diabetic. Method: HPDLC were cultured with AGEs. Expression of MMP-3 and TIMP-1 were determined by immunoflu- orescence and sandwich- in ELISA. Result: The expression of MMP-3 were significantly induced by AGEs. However, AGEs had no effect on the expression of TIMP-1. Conclusion: The resuhs suggest that AGEs may affect ECM metabolism depend- ing on the functional role of MMP-3.
