目的:探讨缺氧诱导因子-1α(HIF-1α)及血管内皮生长因子-C(VEGF-C)在宫颈癌组织中的表达,分析两者与患者年龄、临床分期和预后间的关系。方法:原位杂交组织化学法(ISHH)检测42例宫颈癌组织中HIF-1α mRNA水平,用免疫组化法检测VEGF-C水平。结果:0~Ⅰ期患者HIF-1α mRNA的表达低于Ⅱ~Ⅳ期患者(χ2=12.017,P<0.05),HIF-1α mRNA的表达随着临床分期的增高表达增强,而与年龄无关(χ2=0.001,P>0.05),与预后相关(Zc=2.378,P<0.05)。VEGF-C表达与患者年龄(χ2=1.117)、临床分期(χ2=1.840)、预后(Zc=1.187)均无关(均P>0.05)。VEGF-C、HIF-1α mRNA表达阳性组微血管密度均高于阴性组(Zc分别为4.74和4.40,P<0.01),HIF-1α mRNA表达与VEGF-C表达呈正相关(rs=0.321,P<0.05)。结论:VEGF-C、HIF-1α mRNA在宫颈癌进展过程中可能通过调节肿瘤相关的血管生成起协同作用。
Objective: To investigate the expression of hypoxia-inducible factor-1α (HIF-1α)and vascular endothelial growth factor C (VEGF-C) in cervical tissue and their correlation with the age, clinical stage, and prognosis of cervical cancer. Methods:The expression of HIF-lo~ mRNA in 42 cases of cervical cancer was assessed by in situ hybridization histochem- istry(ISHH). The expression of VEGF-C was assessed by immunohistochemical method. The correlation between expression of VEGF-C with the clinical stage, pathological grade, and prognosis of cervical cancer were analysed. Results:HIF-1α mRNA was over expressed in 26 cases of cervical cancer (61.9%).The expression of HIF-lc~ mRNA was elevated with increased clinical stage (X2= 12.017, P 〈 0.05). There was no association between HIF-1α mRNA expression and age (x2=O.O01, P 〉 0.05). The HIF-1α mRNA levels showed correlation with prognosis (Zc=2.378, P 〈 0.05).There was no association between VEGF-C expression and age(x2=1. 117), clinical stage(x2=1.840) and prognosis (Zc=1. 187, P 〉 0.05,respectively). It was found that MVD was significantly higher in the positive group of VEGF-C and HIF-lc~ mRNA expression (Zc=4.74 and Z,=4.40, P 〈 0.01, re- spectively). There was correlation between the expression of HIF-1α mRNA and VEGF-C (r,=0.321,P 〈 0.05). Conclusion: VEGF-C and HIF-lot mRNA may play an important role in the progression of cervical cancer.
