摘要
为了实现新城疫病毒血凝素-神经氨酸酶(HN)基因在中国仓鼠卵巢细胞中的稳定表达,构建了同时含有HN基因和二氢叶酸还原酶(dhfr)基因的真核表达载体pCI-HN-D。将纯化后的重组质粒通过脂质体转染二氢叶酸还原酶缺陷型中国仓鼠卵巢(CHO/dhf)细胞,经3轮氨甲喋呤加压筛选,获得了稳定表达HN基因的细胞株。用间接免疫荧光和免疫印迹检测目的蛋白的表达情况。结果表明,HN基因在CHO/dhfr-细胞中成功表达。
