抗牛病毒性腹泻病毒单克隆抗体的制备及其特性研究 被引量:5

摘要 利用牛病毒性腹泻病毒(BVDV)基因疫苗和重组E2蛋白分别免疫BALB/c小鼠,制备了5株单克隆抗体(4G3、3B2、3F8、2H6、3A11),并对这些单克隆抗体的亚类、免疫原性及其与瘟病毒的反应性进行了检测。结果显示,4G3、3B2、3F8、2H6为IgM类抗体,3A11为IgG类抗体;纯化后的单克隆抗体与纯化前比较,与抗原的反应性略有提高;5株单克隆抗体与BVDV和猪瘟病毒(CSFV)均有交叉反应性。表明,制备的5株单克隆抗体所针对的是BVDV和CSFV共有的抗原表位。 To provide the effective method for diagnosing bovine viral diarrhea virus(BVDV) and classical swine fever virus(CSFV), BALB/c mice were immunized with gene vaccines(plR-BE2 and plR-BE2m) and recombinant E2 protein of BVDV to prepare monoclonal antibody(McAb) against BVDV,respectively. Then,the prepared McAbs were identified on subtype,immunogenieity and reactivity with antigen, respectively. Five strains of McAbs(4G3,3B2,3F8,2H6, and 3All ) were obtained, of which 4G3,3B2,3F8, and 2H6 were IgM subtype and the 3All was IgG subtype. The reactivity with antigen of purified McAbs were slightly higher than that of non-purified ones. The five McAbs were all cross-reactivatied with BVDV and CSFV. The results manifested the five MeAbs had common epitope against BVDV and CSFV.
