硝酸镧对MDCC-MSB1细胞形态及细胞周期的影响 被引量:1

摘要 以体外培养的MDCC-MSB1细胞为研究对象,分别经终浓度为0、8、32、64μg/mL的硝酸镧作用24、48、72 h后,采用台盼蓝染色测定细胞活性,荧光显微镜观察细胞的形态学变化,流式细胞术测定细胞周期。研究了硝酸镧对鸡MD肿瘤细胞株MDCC-MSB1细胞形态及细胞周期的影响,进而探讨硝酸镧对MDCC-MSB1细胞的影响机制。结果表明,硝酸镧能够降低MDCC-MSB1的细胞活性,能够引起典型的凋亡形态学变化及细胞周期的改变,并呈现细胞凋亡的细胞型,变化均呈现时间-剂量效应。证实,一定浓度的硝酸镧具有诱导体外培养MDCC-MSB1细胞凋亡的作用。 Effect of lanthanum nitrate La(NO3)3 on the cell morphogenesis and cell cycle of Marek's disease(MD) cancer cell Iine(MDCC-MSB1) was studied in vitro to explore its mechanisms. MDCC-MSB1 cells were treated respectively with La(NO3 )3 at 0,8,32 and 64μg/mL for 24,48 and 72 hours respectively. Cell activity were examined by trypan blue staining,morphological changes were examined by fluorescence microscope,and cell cycle were measured by flow cytometry. Cell activity of the MDCC-MSB1 was de- creased, typical apoptosis characteristics were observed by fluorescence microscope and cell cycle was changed,which indicated by time- and dose-response. These results showed that proper concentration of La(NO3)3 could induce the apoptosis of MDC-MSB1 cells cultured in vitro.
