摘要 液体芯片飞行时间质谱技术(ClinProt系统)是近年开发的蛋白质组学新技术,其利用纳米磁珠俘获肿瘤患者与对照者体液中低丰度或低相对分子质量特异蛋白/多肽,经基质辅助激光解吸电离飞行时问质谱测定和软件分析,建立由两者差异表达蛋白/多肽组成的质谱图模型,用于预测未知样品的归属。该系统具有良好的特异性、灵敏性和重复性,高通量,操作简单、快捷等优点,能对所获得的差异蛋白/多肽进行蛋白鉴定,是极具潜力的临床肿瘤早期诊断工具。本文将就目前ClinProt系统在临床肿瘤研究中的应用进行综述。 Liquid chip time-of-flight mass spectrometry technology (ClinProt system) is a new proteomic technology exploited in recent years. Nano-magnetic beads (liquid chip) are used to capture low- abundant or low-mass specific proteins/polypeptides in body fluids of both tumor patients and controls. After MALDI-TOF MS measurements and bioinformatics software analysis, differentially expressed proteins or polypeptides are employed to screen a tumor biomarker pattern, thus unknown samples can be correctly distinguished. ClinProt system is a very promising tool for early diagnosis of tumors due to high specificity, sensitivity and reproducibility, high throughput, easy and simple manipulation. This review is about the latest research and application of ClinProt system in the study of tumor clinical research.
