糖皮质激素对动脉内皮细胞11β-羟基类固醇脱氢酶基因表达的影响

摘要 目的:探讨高浓度葡萄糖刺激下糖皮质激素对大动脉内皮细胞11β羟基类固醇脱氢酶(11β-HSD)基因表达的影响。方法:将猪髂动脉内皮细胞培养于高糖(30mmol/L)中,分别加入不同浓度糖皮质激素刺激,用RT-PCR法检测11β-HSD1,11β-HSD2 mRNA表达情况,Western-blot方法检测11β-HSD1,11β-HSD2蛋白表达情况。结果:正常对照组中,仅有少量的11β-HSD1 mRNA及蛋白表达,高糖状态下,11β-HSD1 mRNA和蛋白表达升高,且随着可的松浓度的增加而增加(P<0.05);高糖状态下,11β-HSD2 mRNA和蛋白表达无明显变化。结论:糖皮质激素使高糖培养下的动脉内皮细胞11β-HSD1表达增加,可能与糖尿病血管病变的发生发展有关。 Objective:To investigate the effect of glucocorticoid on the expression of 11β-hydroxysteroid dehydrogenase in the pig iliac artery endothelial cell (PIEC) under hyperglycemia expression. Methods:PIEC were incubated at a different concentration of glucocorticoid under hyperglycemia(30 mmol/L),the expression of 11β-HSD1, 11β-HSD2 were measured by RT-PCR technique, the protain expression of 11β-HSD1, 11β-HSD2 were measured by Western-blot technique. Results:Under hyperglycemia, glucocorticoid can upregulate the expression of 11β-HSD1 mRNA and protein (P 〈 0.05) compared with the control group,and increased with the concertration of cortisone. The expression of 11β-HSD2 mRNA and protein had not change with the concentration of cortisol. Conclusion: High glucose and glucocorticoid can increase the exoression of 11β-HSD1 mRNA and protein.
