高胆固醇喂养ApoE基因缺陷小鼠的心脏改变

摘要 目的动态观察高胆固醇喂养的不同周龄ApoE基因缺陷小鼠心脏形态学以及心肌胶原的变化。方法以24、32周和40周龄ApoE基因缺陷鼠(n=6/组)为研究对象,相同周龄的C57BL/6J小鼠设为对照。常规检测血清胆固醇水平;HE染色观察各组小鼠心脏形态学改变,Masson染色观察心肌胶原改变,KS400图像分析系统测量心肌胶原容积分数(CVF)和心肌血管周围胶原面积和管腔面积之比(PVCA),苦味酸天狼猩红染色进行左室胶原特异染色及定量分析,观察Ⅰ型和Ⅲ型胶原的变化。结果40周龄ApoE-/-小鼠左心室前壁、左心室侧壁、左心室后壁较24周龄ApoE-/-小鼠增厚(P<0.05);24、32和40周龄ApoE-/-小鼠随着周龄增加心肌胶原含量随之增加(P<0.05),Ⅰ型胶原逐渐增多。结论高胆固醇喂养的ApoE基因缺陷小鼠,随着胆固醇水平增加,心脏增大,心肌胶原含量显著增加。 Objective To study the dynamic changes in cardiac morphology and myocardial collagen in apollpoprotein E-deficient (apoE-/-) mice of different age with cholesterol fed. Methods 18 male apoE-/- mice were randomized into 3 groups: 24 weeks group (n=6), 32 weeks group (n=6), 40 weeks group (n=6). 8 C57BL/6J mice were set as the control. Serum cholesterol, low-density lipoprotein cholesterol (LDL-C), triglyceride, high-density lipoprotein cholesterol (HDL-C) were determined. The cardiac morpho logical changes and the myocardial collagen were observed. Results Compared with the 24 weeks group, the content of myocardial collagen of 32 and 40 weeks group significantly increased (P〈0.05). Conclusion With the increasing of age, TC significantly in creased, myocardial collagen content tended to go up.
