硒对砷染毒致雄性大鼠生殖损害的拮抗作用 被引量:4

摘要 目的探讨硒对高砷所致雄性大鼠生殖损害的拮抗作用。方法50只雄性Wistar大鼠随机分成对照组、砷(As2O3150mg/L)组、砷加低硒(0.5mg/L亚硒酸钠)组、砷(As2O3150mg/L)加硒(2.0mg/L亚硒酸钠)组、砷(As2O3150mg/L)加高硒(4.0mg/L亚硒酸钠)组5组,通过饮水加砷、加硒的方法染毒10周。观察砷、硒对大鼠体重、睾丸和附睾质量及其脏器系数,精子计数、活动率及畸形率,睾丸、附睾生化标志酶,血清性激素,睾丸组织病理切片以及初级精母细胞染色体畸形率的影响。结果①右侧睾丸质量对照组与砷加高硒组相比差异有统计学意义(P<0.05)。②砷组精子计数、精子畸形率、精子活动率、乳酸脱氢酶、睾丸酮和促黄体生成激素与对照组比较差异均有统计学意义(P<0.05)。③睾丸组织病理学检查结果显示,砷组与对照组比较结构较疏松,砷加低、中、高硒组与砷组比较结构相对完整。④初级精母细胞染色体畸形率结果显示,4组染毒组与对照组比较差异无统计学意义(P>0.05)。结论硒可拮抗高砷的雄性生殖毒性,以中等剂量硒效果最好。 Objective To study the antagonism of selenium on male reproductive toxicity induced by excessive arsenic. Methods 50 male Wistar rats were randomly divided into five groups: control group, arsenic group ,arsenic plus low dose selenium, arsenic plus mid-dose selenium and arsenic plus high-dose selenium groups. After the animals were fed with deionized water supplemented with As203 or/and Na2SeO3 for 10 weeks,the body, right testical, right epididymitis and its organic coefficient were measured and calculated, the count of sperm and the rates of sperm mobility and aberration were analyzed, the levels of lactate dehydrogenase (LDH), serum testosterone (T) and luteinizing hormone (LH) were examined, meanwhile the pathological al- terations of test and the chromosome aberration of primary spermatocytes were observed. Results ① Weight of right testical was significantly reduced ( P 〈 0.05 ) in arsenic plus high-dose selenium group as compared to control group. ②The sperm count, the aberration rate of sperm, the rate of sperm mobility, the levels of LDH and T in serum, and the LH level were significantly changed in arsenic groups as compared to control group respectirely.③ The microstructure of testis was loosened in arsenic group as compared to control rats. Compared with arsenic group, low, middle, and high doses of selenium could tion of testis induced by arsenic. signifi④No significant difference ( P 〉 0.05 ) candy alleviate the pathological aherawas observed after arsenic and/or selenium treatments in the rate of chromosome aberration of primary spermatocytes as compared to control group. Conclusion Arsenic can result in reproductive toxicity in mate rats, and appropriate selenium take can antagonize the toxicity of arsenic.
