摘要
根据Genbank中禽多杀性巴氏杆菌的omp(outer membrane protein)基因核苷酸序列设计1对特异性引物,应用PCR方法扩增出禽多杀性巴氏杆菌(5∶A)C48-1株的omp基因全长片段,与克隆载体pmD18-T连接后,转化感受态细胞,对经PCR鉴定和酶切鉴定均为阳性的克隆进行测序。结果表明omp基因全长2 376bp,编码791个氨基酸。序列分析表明该菌株与PBA100株、P52株、EU570212、PM70株核苷酸同源性为48.7%~98.6%,氨基酸同源性为34.8%~99.0%。
A pair of primers was designed according to the sequence of outer membrane protein (omp) gene of Pasteurella multocida published in GenBank. Full-length omp gene of avian P. multocida (5 : A) C48 - 1 strain was amplified by PCR using the primers and cloned onto proD18 - T vector. The recombinant plasmid was identified by using digested restriction endonuclease and PCR. The result showed that the length of the nucleotide of omp gene was 2376 bp encoding 791 amino acids. The C48 -1 strain had 48.7% to 98.6% nucleotide sequence homologies on omp gene, and 34.8% to 99.0% deduced amino acid homologies, as compared to P. multocida PBA100, P52, EU570212 and PM70 strains.
