摘要
目的:观察EB病毒BHRF1反义寡核苷酸片段对鼻咽癌SUNE1细胞株增殖的影响。方法:PCR、3HTdR的掺入法及电泳等。结果:PCR从SUNE1细胞DNA中扩增出EB病毒的4种(BHRF1、DNA酶、胸苷激酶及核抗原1)全基因片段;适当浓度BHRF1反义寡核苷酸片段能引起无血清培养的SUNE1细胞株增殖受抑制;透射电镜及琼脂糖凝胶电泳检测结果发现与SUNE1细胞发生凋亡有关。结论:适当浓度BHRF1反义寡核苷酸片段能抑制无血清培养的SUNE1细胞株的增殖。
Objective:To investigate the effects of EBV BHRF1 antisense oligodeoxynucleotide (ASO) on the proliferation of poorly differentiated nasopharyngeal carcinoma (SUNE1) cell lines. Methods:Polymerase chain reaction (PCR),3HTdR incorporation and electrophoresis. Results:Intact BHRF1, DNase, thymidine kinase (TK) and nuclear antigen1 gene fragments of EBV were amplified using PCR technique with DNAs from B958 and SUNE1 cell lines as template. Inhibited proliferation of SUNE1 was observed in the absence of fetal calf serum(FCS) and exposed to BHRF1ASO. The results from electron microscopy and agarose gel electrophoresis indicated that apoptosis appeared in SUNE[JX*1/9]1 cells. Conclusion:Absence of FCS and exposed to BHRF1ASO on proper concentratin could inhibit proliferation of SUNE1.
