摘要
为探究芍药胚苗快繁的合适培养基,在初步明确6-BA用量为0.5 mg.L-1的前提下,采用二次正交旋转组合设计法探讨3个因素(赤霉素、蔗糖、葡萄糖)对芍药胚苗快繁的影响。结果表明:在试验浓度范围内,葡萄糖用量对腋芽诱导的影响最大,其次是赤霉素和蔗糖,且葡萄糖和蔗糖的用量存在互作效应。芍药腋芽诱导3因子的最优组合是:赤霉素为1.84 mg.L-1、蔗糖为11.02 g.L-1、葡萄糖为17.13 g.L-1,在该组合的培养基上,腋芽的诱导系数在接种40 d时可以达到理论最大值3.05。
To explore the suitable medium for axillary bud induction from Herbaceous Peony seedling, the effects of the three factors, GA3, sucrose and glucose were studied by using of orthogonal and rotational combination design based on the primary result that 0.5 mg.L^-1 6-BA applied to MS medium was suitable for Peony axillary bud induction. The research results revealed that: in this experiment range, the glucose content had the most important influence on axillary bud induction and it was followed by GA3 and sucrose. Interaction existed between the effects of sucrose and glucose on axillary bud induction. The optimum combination of these three factors for axillary bud induction was GA3 1.84 mg.L^-1 , sucrose 11.02 g.L^-1 and glucose 17.13 g.L^-1. The highest axillary bud induction ratio was 3.05 in theory after 40 days cultured on this medium.
