摘要 目的观察大鼠甲状旁腺素1-34(rat parathyroid hormone1-34,rPTH1-34)对体外培养新生大鼠心室肌细胞活力的影响。方法原代培养新生大鼠心室肌细胞,以0.1μmol/LPTH1-34分别孵育0、12、24、48、72h,噻唑蓝(MTT)比色法测定细胞活力。结果心肌细胞活力在12h时最大(P<0.01),24h时开始下降,但仍高于正常水平(P<0.01),48h时降至正常水平,到72h降至正常水平以下(P<0.05)。结论0.1μmol/LPTH刺激12h时心肌细胞活力最旺盛。 Objective Aim To investigate effect of rat parathyroid hormone 1 -34 on cardiac myocyte viability in neonatal rat. Methods Using primary cultured neonatal ventricular cells of Wistar rats, the cardiomyocyte viability was measured with MTT method after irritating with 0. 1 μmol/L PTH1 - 34 at different time points including 0, 12, 24, 48 and 72 h. Results The maximum value of myocyte viability presented at 12 h (P 〈0. 01). The value was back to baseline at 48 h. At 72 h, the adherence capability of cardiomyocytes decreases under normal level. Conclusion The maximum value of myocyte viability presented at 0. 1 μmol/L PTH in 12 h.
