摘要
目的探讨黄芪对2型糖尿病大鼠心肌基质金属蛋白酶-9(MMP-9)表达及活性的影响。方法将SD大鼠36只随机分为正常对照组、2型糖尿病模型组和黄芪治疗组(黄芪8 g/kg)。各组分别给药12周后,显微镜观察其心肌结构,采用苦味酸-甲苯胺蓝染色、免疫组化和明胶酶图的方法检测心肌组织中胶原(CL)含量、MMP-9的蛋白表达和活性水平。结果模型组大鼠心肌纤维局灶性坏死、细胞外基质堆积,心肌CL含量、MMP-9的蛋白表达和活性水平均高于正常对照组,黄芪治疗组以上病理改变较模型组减轻。结论黄芪可通过改善全身代谢及降低心肌MMP-9的蛋白表达、活性水平和CL含量等途径来发挥其在2型糖尿病中的心脏保护作用。
Objective To investigate the influence of Radix Astragali seu Hedysari on expression and activity of matrix metallopmteinase-9 (MMP-9) of myocardium in rats with type 2 diabetes. Methods All SD rats (n = 36) were randomly divided into the normal control group, model group of type 2 diabetes and treatment group (with 8 g/kg of Radix Astragali seu Hedysari). After 12 weeks the structure of myocardium was observed by the microscope, and the content of collagen (CL), expression and activity of MMP-9 in myocardium were detected by picric acid-toluidine blue staining, immunohistochemical assay and gelatin zymography. Results In the model group of type 2 diabetes the focal necrosis of cardiac muscle fibers and the accumulation of extracellular matrix were observed, and CL content, expression and activity of MMP-9 in myocardium were all higher than those in the normal control group. In the treatment group these pathological changes were alleviated than those in the model group. Conclusion Radix Astragali seu Hedysari can protect the heart in patients with type 2 diabetes through promoting whole body's metabolism, reducing the expression and activity of MMP-9 and CL content in myocardium.
