鱼腥草素衍生物对TNF-α诱导的大鼠血管平滑肌细胞Syndecan-4表达的影响 被引量:1

摘要 目的观察一种鱼腥草素衍生物(houttuyfonate derivative,HD)对肿瘤坏死因子α(tumor necrosis factorα,TNF-α)诱导的大鼠血管平滑肌细胞(vascular smooth muscle cells,VSMCs)增殖及Syndecan-4表达的影响。方法以终浓度分别为TNF-α20ng/mL、HD 4μg/mL、HD 8μg/mL、TNF-α20 ng/mL+HD 4μg/mL及TNF-α20 ng/mL+HD 8μg/mL对体外培养的VSMCs作用24 h,并设立相关对照组,用MTS/PMS法检测VSMCs的增殖,用Western blot蛋白免疫印迹法测定Syndecan-4的表达情况。结果与对照组相比,HD各剂量组单独应用对VSMCs的增殖无明显作用(P>0.05);HD 4μg/mL和8μg/mL可分别抑制TNF-α对VSMCs的增殖作用(P<0.05)。Western blot测定显示:与对照组相比,HD各剂量组单独应用对Syndecan-4的表达无明显影响(P>0.05),HD 4μg/mL和8μg/mL均可抑制TNF-α刺激后Syndecan-4的表达(P<0.05)。结论HD对TNF-α刺激后的VSMCs的增殖及Syndecan-4表达均有抑制作用。 Objective To investigate the effects of a new houttuyfonate derivative(HD) on proliferation of VSMCs and expression of Syndecan-4 induced by TNF-α in vitro.Methods Rat VSMCs were cultured and exposed to TNF-α or HD respectively,and then cotreated with TNF-α and HD.All the groups were cultured for 24 hours in vitro,in addition to the untreated control group established for comparison.The ratio of proliferation of VSMCs was determined by non-radioactive MTS/PMS assay and the expression of Syndecan-4 was evaluated by western blot using anti-Syndecan-4 antibody.Results Statistical analysis showed that,compared to the control group,HD alone had no effect on VSMCs growth,but significantly inhibited VSMCs proliferation induced by TNF-α.It also showed that compared to control group,HD alone had no effect on the expression of Syndecan-4,but significantly inhibited the expression induced by TNF-α(P〈0.05).Conclusions HD can inhibit proliferation of rat VSMCs and expression of syndean-4 protein induced by TNF-α in vitro.
