摘要
研究了细菌阿特拉津氯水解酶基因atzA在转基因水稻AA269株系自交和回交后代中的遗传规律.结果表明:atzA基因作为一个显性遗传位点遵循孟德尔遗传分离规律.除草剂抗性鉴定结果表明,在T_1代水稻群体中除草剂抗性和敏感株数遵循3:1的分离规律,T_2代稳定纯合;T_2~T_7自交世代的转基因植株均保持稳定的除草剂抗性.在BC_1~BC_3代群体中除草剂抗性和敏感株数遵循1:1的分离规律,BC群体自交的BCF_2群体中遵循3:1分离规律.分别对AA269/9311的B_3F_1和B_2F_2群体中的一个株系进行了PCR分析,PCR阳性株与阴性株的分离比例分别符合1:1和3:1,与除草剂抗性鉴定结果一致.对T_7代群体中部分单株的Southern杂交和RT-PCR分析表明,atzA基因已稳定遗传至T_7代,并得到表达.
The genetic rule of bacterial atrazine chlorohydrolase gene atzA in self-cross and back-cross progenies of transgenic rice "AA269" were investigated. The results showed that as a dominant genetic locus,atzA gene followed Mendel's genetic segregation rule. The herbicideresistance identification results showed that herbicide-resistance plants and herbicide-sensitivity plants followed 3:1 segregation rule in T1,and stably homozygotic in T2 and herbicide-resistance had been stably inherited to T7,and BC1-BC3 and BCF2 (self-cross of BC)also followed 1:1 and 3:1 segregation rules respectively. The segregation ratio of PCR positive plants and PCR negative plants of BC3F1 and BC2F2 followed 1 : 1 and 3 : 1 segregation rule. The PCR results are identical to that of herbicide-resistance identification. The Southern blot hybridization and RTPCR analysis showed that atzA gene had been stably inherited to T7 generation and expressed steadily in T7.
