摘要 食品添加剂红曲红中桔霉素限量问题已成为近年来国内外研究热点。目前,国内缺乏专门针对红曲红色素中桔霉素检测的标准方法。文中在综合分析国内外相关桔霉素检测方法技术资料的基础上,通过试验,对粉状样品预处理条件进行了优化,研究了适合我国生产的红曲红色素中桔霉素检测方法。粉状红曲红样品先用体积分数50%乙醇配制成一定浓度的试样液,试样液在优化的树脂分离条件下进行洗脱,然后采用高效液相色谱法测定洗脱液中的桔霉素含量。实验结果显示:优化的树脂分离条件能较好地洗脱样品中的桔霉素;配有荧光检测器的高效液相色谱仪能较灵敏地检出桔霉素含量,方法检测限为0.2μg/L,基质加标平均回收率为105.5%,相对标准偏差(RSD)为2.2%。 Limited usage of citrinin in food additive monascus color has drawn worldwide attention recently. There is no standard method for the determination of citrinin content in domestic monascus color. In this study, the pretreatment conditions of the test sample were optimized and the standard method for the determination of citrinin content in domestic monascus color was provided by studying and analyzing related research both domestic and abroad. The powdered monascus color sample was dissolved in 50% ethanol-water mixture. Then citricin in the eluted solution was detected by HPLC with Spectrophotofluorometer. The test results showed that the citricin in the sample was fully washed into the eluted solution on the optimized pretreatment conditions. The detection limlit for citrinin content in domestic monascus color was 0.2μg/L, the mean recovery rate of matrix spiked samples was 105.5%, and the relative standard deviation was 2.2%.
