摘要
采用有机溶剂回流法提取和大孔树脂纯化龙舌兰总皂苷,并用正交实验确定最佳提取工艺。结果显示:乙醇提取法最佳工艺为,采用体积分数65%的乙醇,料液比(g:mL)1∶14,90℃回流提取2h,重复2次。D101型大孔树脂对纯化工艺为以体积分数70%的乙醇为洗脱剂,5倍柱体积洗脱,皂苷平均纯度达到75.3%。经薄层层析分析显示龙舌兰总皂苷中含有3种皂苷,其中1种为人参皂苷。
This article discussed the techniques of extracting and purifying total saponin with organic solvent and macroporous resin from the maguey. The optimum technical condition of extracting maguey saponin was decided by orthogonal design. The result showed that the extraction condition of 65% alcohol, 1 : 14 proportion of material and solvent, extracting for 2h at 90 ℃ was the optimum condition. The D101 macroporous resin was used to purify the saponin at the condition using 70% ethanol as the eluting solvent, 5 times of columns volumes. The average purity of saponin achieves 75.3%. Three kinds of saponin were analyzed by thin layer chromatography and ginseng saponin was one of them.
