受体血浆浸泡+液氮冷冻同种异体血管移植的实验研究 被引量:1

摘要 目的观察受体血浆浸泡+液氮冷冻处理同种异体血管抗原性、移植后免疫排斥反应及组织结构变化。方法对新鲜家兔股动脉分别行液氮冷冻及受体血浆浸泡+液氮冷冻处理,测定MHC-Ⅰ类抗原含量。30只家兔随机分为3组,每组10只。A组:移植新鲜异体血管;B组:移植液氮冷冻血管;C组:移植受体血浆浸泡+液氮冷冻血管。在移植后2、8、16周测定各组体外微量淋巴细胞数量及在6、12周观察血管组织结构变化及血管通畅率变化。结果C组MHC-I类抗原表达量水平明显下降(P<0.05);C组淋巴细胞死亡率明显低于B组(P<0.05);光镜观察C组术后6周管腔有内皮层覆盖,12周中膜出现平滑肌层。结论受体血浆浸泡+液氮冷冻处理同种异体血管可使抗原性进一步下降。移植后体外微量细胞毒实验及组织学观察发现免疫排斥反应不明显。 Objective To observe the antigenicity,graft rejection and changes in organization after treated with recipient plasma immersion+frozen in liquid nitrogen.Methods The fresh rabbit femoral artery was treated with frozen in liquid nitrogen and immersed in the recipient blood plasma+frozen in liquid nitrogen,respectively.Determinate MHC-ⅠandⅡ antigen content,and compare with fresh femoral artery(control group).Thirty rabbits were randomly divided into 3 groups:group A was graft with fresh femoral artery of rabbit,group B was graft with preserved with liquid nitrogen,and group C was graft with preserved by liquid nitrogen+immersed by recipient blood plasma of rabbit.The study of lymphocy toxicity were determined at 2 weeks,8 weeks and 16 weeks after operation.The histologic change and inmunologic property was observed at 6 weeks and 12 weeks after operation.Results The MHC-I and Ⅱ antigenicity of group C decreased obviously compared with either group A or group B(P〈0.05).The contents of lymphocy toxicity of group C had significant difference to group B(P〈0.05).The patency rate of groups was no significant difference.Endothelium was seen in 6 weeks after operation and smooth muscle cells were seen in 12 weeks after operation.Conclusion Immersed with recipient blood plasma before reserved with liquid nitrogen could obviously decrease the antigenicity and the transplant effect was better than the only liquid nitrogen group.
