摘要
目的:建立了LC-APCI-MS/MS法测定全血中苯妥英钠的血药浓度。方法:采用Zorbax Eclipse XDB-C18(4.6×150 mm,5μm)色谱柱,流动相为乙腈-0.1%甲酸溶液(50:50)。大气压喷雾离子源(APCI),正离子检测,采用多反应监测方式进行定量分析,检测离子为m/z 253→182(苯妥英钠),m/z 316→270(内标氯硝西泮)。结果:全血中苯妥英钠浓度在4~2400 ng/ml范围内线性关系良好,最低检测限为2 ng/ml。苯妥英钠的萃取平均回收率为71.45%~87.27%,日内及日间RSD都低于6%。结论:该法操作简便、快速、灵敏度高,可用于苯妥英钠的临床毒物分析。
Objective:To establish a LC-APCI-MS/MS method for determination of phenytoin sodium in blood.Methods:A Zorbax Eclipse XDB-C18(4.6×150 mm,5 μm) column was used,with mobile phase of acetonitrile-0.1% formic acid(50:50).Atmospheric pressure chemical ionization(APCI)source was applied and operated in positive ion mode.Multiple reaction monitoring(MRM)mode was used.The detected ions were m/z 253→182(phenytoin sodium),m/z 316→270(internal standard,clonazepam).Results:The calibration curve of phenytoin sodium was linear in the concentration range of 4~2400 ng/ml.The LOD was 2 ng/ml.The average extraction recovery was 71.45%~87.27%.The RSDs of inter-day and intra-day were less than 6%.Conclusion:The analytical method for phenytoin sodium is accurate,sensitive and suitable for clinical toxicological analysis.
