荧光实时PCR检测法在突发公共卫生事件的应用 被引量:8

摘要 目的:应用实时荧光PCR(real-time fluorescence PCR)方法及国标(GB)方法,对东莞市2006~2007年突发公共卫生事件中可疑食品及生物样品,检测食源性病原体,对两种方法的检出率及效率进行比较。方法:针对2006~2007年东莞市突发公共卫生事件,同时运用实时荧光PCR方法及国标方法对沙门菌(Sa)、副溶血性弧菌(Vp)、金黄色葡萄球菌(Sta)及志贺菌(Sg)等病原菌进行增菌、分离及鉴定,并将两者结果进行比较。结果:实时荧光PCR与GB两种检测方法对突发公卫事件中所涉及样品的目标病原体检出具有较好的一致性,两种检测方法对肛拭及粪便样检出率,实时荧光PCR方法要比GB法高,对Vp、Sg及Sa的检出也比GB法高。结论:实时荧光PCR与国标检测方法组成实验方案,准确、快速、简便、特异性强,为其应用于突发公共卫生事件中可疑食源性病原体检测奠定良好基础。 Objective:Utilizing real-time fluorescence PCR and national standard method adopted at present to detect food relate disease outbreak of public health emergency that has taken place in DongGuan city during 2006~2007,Then compare the fact of efficient with the two methods.Methods:Discuses outbreak of public health emergency in Dongguan city during 2006~2007,To application method of multi-link real time fluorescence PCR for detect Pathogens such as Shigella Salmonella Staphylococcus aureus and Vibrio parahaemolyticus in foods and others biological samples.At the same time application national standard math to handle with those sample.Then compare the result of the two methods in efficient and accurate.Results:In outbreak of public health emergency the two methods were used to detect objective pathogen has preferably coherence,positive rate in detection anal swab sample and feces sample,real-time fluorescence PCR is higher than GB,positive rate in detection objective pathogen such as Vp,Sg and Sa real-time fluorescence PCR is higher than GB.Conclusion:The scheme of combination experiment consists of real-time fluorescence PCR and national standard method rapid more accurate and purposeful.Make good basic for Application to detect food relate disease outbreak of public health emergency.
