燕麦生物碱提取物的抗氧化与降血脂作用研究 被引量:26

摘要 研究燕麦麸皮生物碱提取物(OBAE)对D-半乳糖诱导衰老小鼠血脂代谢和氧化应激状态的影响及其作用机理。将健康的昆明雄小鼠随机分为对照组、模型组、和低、中、高OBAE 3个剂量组。饲养7周后,测定不同剂量OBAE对小鼠血脂水平、抗氧化酶活性、脂质过氧化产物丙二醛含量及脂蛋白脂酶的表达的影响。同时还分析了提取物中生物碱和多酚的含量。结果表明,摄入OBAE后小鼠血脂水平降低,血清高密度脂蛋白增加,丙二醛的含量减少,而血清抗氧化酶活性显著增加,脂蛋白脂酶mRNA表达显著上调。对OBAE的主要抗氧化成分进行分析,燕麦生物碱的含量:627 mg/kg,总多酚的含量509 mg/kg。燕麦麸皮生物碱提取物(OBAE)具有抗氧化、降血脂生物活性,所含的燕麦生物碱、多酚是其发挥作用的主要物质基础。 By using D-galactose induced aged mice,the effect and mechanism of oat bran avenanthramide extract(OBAE) on oxidation and hypolipidemic for mice were investigated.Male KM mice(60) were divided into five groups and were fed with normal diet(control and model groups),diets added with OBAE of 250 mg,500 mg, and 1 000 mg/kg,ig respectively(three dose groups) for seven weeks.Then,the serum lipid profile,the antioxidation enzyme activity and the malondialdehyde content of the mice were determined,and the LPL mRNA expression was also evaluated.Results: To the aged mice,feeding OBAE significantly reduces serum malondialdehyde amount,and enhances the activities of SOD and GSH-px in serum.The levels of serum TC and TG of the mice fed with OBAE significantly reduce,while the HDL-C increases.OBAE can significantly enhance the LPL mRNA expression.The contents of avenanthramides and total phenols in the OBAE were measured as 627 mg/kg and 509 mg/kg,respectively.It is demonstrated that OBAE has remarkable antioxidation and hypolipidemic effects on aged mice,which might result from the avenanthramides and total phenols in it.
