酵母RNA的药效学研究

摘要 目的:探讨酵母RNA的肝脏保护、免疫调节以及延缓衰老的作用。方法:①小鼠四氯化碳腹腔注射,建立急性肝损伤模型,行ALT、AST检测及肝组织学检查;②用5-氟尿嘧啶建立免疫抑制模型,行巨噬细胞吞噬率、血清TNF-α、IL-2检测,计算脾脏指数、胸腺指数;③30只22月龄Wister大鼠随机分为正常组、酵母RNA组和VitE组,检测脑匀浆的SOD、MDA。结果:酵母RNA组小鼠血清ALT、AST明显低于模型组(P<0.01~P<0.05),肝组织中炎症细胞浸润、肝细胞坏死和变性损害也显著轻于模型组;与5-氟尿嘧啶组相比,酵母RNA可显著提高巨噬细胞吞噬率、脾脏指数、胸腺指数(P<0.01)、血清TNF-α和IL-2(P<0.05);和正常组相比,酵母RNA可显著提高老年大鼠脑匀浆的SOD,降低MDA(P<0.01)。结论:酵母RNA对急性肝损伤有一定保护作用,对免疫低下小鼠有免疫增强作用,对老年大鼠有延缓衰老的作用。 Objective: To investigate yeast RNA's multifunction on protection agaist liver damage,immunity function and anti-senile. Method: (1)Animal model(mouse) of acute hepatic injury was induced by intraperitoneally injected carbontetrachloride discontinuously. The contents of ALT, AST of the serum were measured and morphological changes of hepatic tissue were watched. (2)Animal model of immune suppression was induced by 5-Fu. The splenic index, thymic index, macrophage phagocytosis rate and the contents of IL-2 ,TNF-α of the serum were measured. (3)Thirty Wister rats of 22 months old were randomly divided into three groups: normal group ,yeast RNA group and VitE group. The contents of SOD and MDA of the brain homogenate were measured to observe the effect of yeast RNA on anti-senile in old rats. Result: compared with the model group ,yeast RNA could obviously decrease levels of serum ALT and AST ( P 〈 0.01 - P 〈 0.05 ) . Under light microscopy, the infiltration of inflammatory cells, hepatic cell degeneration, and necrosis in the model group were more serious than those in the yeast RNA groups. Compared with the 5-Fu group,yeast RNA could obviously increase the splenic index, thymic index, macrophage phagocytosis rate(P 〈 0.01 )and the contents of IL-2, TNF-α of the blood serum (P 〈 0.05 ). Compared with the normal group, yeast RNA could obviously increase the level of SOD of rat brain homogenate and decrease level of MDA of rat brain homogenate (P 〈 0.01 ). Conclusion: Yeast RNA has multifunction on chemical protection agaist liver damage, regulation immune function of animal model of immune suppression and anti-senile in old rats.
